The Big Sisters are adolescent Little Sisters that have become unstable in Rapture's environment. They have the appearance of a thin, feminine version of Big Daddies, possessing very similar metallic equipment. The Big Sisters wear diving suits reminiscent of the Big Daddies, but they have a distinct aesthetic, since they are built from found items, rather than completely manufactured. A Big Sister helmet consists of a large sphere with a single red "eye". Big Sisters have very agile frames, being described as almost seven feet tall, and very slim. They wield huge, wrist-mounted ADAM needles on their left arms, used primarily for melee attacks and extracting. In addition, there appears to be a gun barrel directly above her bayonet/needle. The Big Sisters also have a basket on their backs which is used to safely carry Little Sisters. They are charged with maintaining the current ecological balance in Rapture.

There is an original Big Sister, preceding the others. It is speculated that she might be Eleanor Lamb.

Years after the events of BioShock, the Big Sisters have been briefly traveling to the surface to abduct little girls all over the world. Individuals from various organizations started to show interest in the abductions, and begin a thorough investigation. One among them, Mark Meltzer, begins tracking patterns in the abductions. After some planning, Mark encounters a Big Sister and tries to capture her by setting a net trap. She then strikes back, breaking Mark's leg and four of his ribs. She then follows Mark home after he is out of the hospital and abducts his daughter, Cindy. She proceeds to abduct children, while still being tracked by Mark, determined to find his daughter.

Big Sisters are one of the main enemies in BioShock 2. According to BioShock 2 creative director Jordan Thomas, after the player invokes the wrath of a Big Sister, there will be a set amount of time before she arrives at the player's location. He stated that once she arrives, the player's going to have "the fight of their life".

As mentioned previously, there will be a set amount of time before the Big Sister arrives to attack the player. This allows them to rig the environment to their advantage, hacking machinery and laying traps before preparing themselves for the fight. From the videos showcasing the Big Sister in action, she will move very fast from place to place, similarly to a Spider Splicer. Her only weapon, besides her Plasmids, is an oversized needle she will stab the player with. As such, movement-impeding plasmids and weapons would be recommended when taking her on, as well as attacks not requiring a great amount of aim. It is possible she will have the same weaknesses as a Big Daddy.

  • The 'needle' attached to the Big Sister's wrist appears to be a rifle's bayonet.
  • There appears to be a gun barrel located beside the Big Sister's 'needle'. At this point, however, this is unclear.
